基本氨基酸有二十个种，但是人体内存在的氨基酸只有18种，称为人体所需的18种氨基酸

18种中以下八种氨基酸人体不能自己制造，需要从食物中摄取，我们称之为必须氨基酸。

此外人体合成精氨酸、组氨酸的能力不足于满足自身的需要，需要同时从食物中摄取，故称为半必需氨基酸
氨基酸含量测定：
一般样本多为蛋白质或者多肽类物质，在测定前要水解成为氨基酸之后才能测定，
一般所测样本都是经盐酸水解成为游离氨基酸后再进行测定，但是酸水解时会破坏色氨酸、胱氨酸、半胱氨酸等几种氨基酸。所经过酸水解的样本可以同时测定苏，丝，谷，脯，甘，丙，缬，蛋，异亮，亮，酪，苯丙，组，赖和精氨酸等16种氨基酸。
需要测定酸水解会破坏的色氨酸、胱氨酸、半胱氨酸这些氨基酸时一般要对样本进行碱水解或者其他水解，再进行测定。
所以一般测定步骤是样本先经酸水解后先测定不被酸分解的氨基酸，然后根据需要再测定酸会破坏的几种氨基酸。
