◆概况
核酸酶P1分解RNA和热变性DNA的3'-5'-磷酸二酯键。另外,核酸酶P1还分解单核寡核苷酸的3'-磷酸单酯键。如果的氯化钠(pH 6.0)含量超过400mmol/L,它不作用于双链DNA。
◆应用实例
A. |
Sample : 200 μL of RNA or heat-denatured DNA at a density of 2 mg/mL. |
B. |
Reaction Buffer : 200 μL of 0.01 mol/L barbital(veronal) acetate buffer, pH 5.3*. |
C. |
Enzyme : 100 μL of 100 units/mL of Nucelase P1 solution with distilled water. |
D. |
top Solution : 500 μL of cold uranyl acetate buffer (0.25% of uranyl acetate and 2.5% perchloric acid) |
*barbital (veronal) acetate buffer, pH 5.3: Prepare 0.01 mol/L Barbital Sodium Salt in water and adjust it to pH 5.3 with acetic acid.
1. Mix each 200 μL of A, B and C.
2. Incubate it at 50 degree C for 1 hour.
3. The reaction can be stopped by adding 500 μL of D.
